Citicoline is a cognitive enhancer and psychostimulant with significant neuroprotective effects. It participates in the synthesis of phosphatidylcholine (an important component of cell membranes) and supports the synthesis and regeneration of nerve membranes. Citicoline can also stimulate synapse formation, inhibit the process of demyelination, and promote myelin regeneration while preventing neuronal apoptosis. Citicoline exists in the human body in the form of CDP choline and is a precursor for the synthesis of phosphatidylcholine.
Researchers have summarized the application research of citicoline in different diseases in recent years. The research methods include animal experiments, clinical trials and pharmacological studies. The diseases involved in the experiments include ischemic stroke, brain trauma, Parkinson's disease, Alzheimer's disease, glaucoma, drug addiction and mental disorders.
1. Ischemic stroke: Citicoline prevents the accumulation of free fatty acids and protects nerve cells from the activation of A2 phospholipase by inhibiting the neuronal apoptosis mechanism associated with cerebral ischemia. It also accelerates nerve cell regeneration, enhances neuroplasticity, and promotes rehabilitation and recovery of consciousness.
2. Brain trauma: Citicoline, as a precursor for membrane phospholipid synthesis, helps neurological function recovery after brain trauma. Clinical studies have shown that intravenous injection of citicoline can improve patients' cognitive and motor functions.
3. Parkinson's disease: Citicoline combined with levodopa can reduce the dose of levodopa, thereby reducing adverse reactions. Citicoline works by increasing dopamine concentrations in the striatum and substantia nigra.
4. Alzheimer's disease: Citicoline improves cognitive parameters, increases cerebral blood flow, and improves brain bioelectric activity. When used in combination with cholinesterase inhibitors, citicoline can enhance acetylcholine concentrations and improve cognitive function test results.
5. Glaucoma: Citicoline has shown the effect of protecting retinal nerve cells and improving visual field parameters in the treatment of glaucoma. It can be administered in the form of intramuscular injection, oral administration, or eye drops.
6. Drug addiction and mental disorders: Citicoline shows positive effects in the treatment of methamphetamine and cocaine addiction, improving cognitive function and reducing drug demand. In the treatment of alcohol withdrawal syndrome, citicoline improves patients' attention and spatial orientation ability.
Metabolism and pharmacological effects of citicoline
The metabolic process of citicoline in the human body includes hydrolysis to choline and cytosine in intestinal epithelial cells. These metabolites can cross the blood-brain barrier and resynthesize CDP choline in the brain. Citicoline and its metabolites are involved in the biosynthesis of acetylcholine, nucleic acids and methionine.
Citicoline is essential for the normal synthesis and reconstruction of nerve membranes and the function of acetylcholine, nucleic acid and protein synthesis pathways.
