Does Nicotinamide Mononucleotide Have a Positive Effect on Health in Older Adults?

A recent study published in the journal GeroScience has unveiled the positive effects of beta-Nicotinamide Mononucleotide (NMN) on the health of the elderly. This double-blind, randomized, placebo-controlled study involved 60 participants aged between 65 and 75, aiming to assess the impact of a 12-week NMN supplementation on blood nicotinamide adenine dinucleotide (NAD+) levels, physical function, particularly walking function, and collected data related to sleep and stress as secondary endpoints.
The study results revealed that, compared to the placebo group, the NMN group showed a significant increase in blood NAD+ and its metabolites after 12 weeks. Moreover, the 4-meter walking time in the NMN group was significantly reduced, indicating the maintenance of walking speed. Notably, the study also found a significant negative correlation between NMN intake and the changes in blood NAD+, N1-methyl-2-pyridone-5-carboxamide (2-PY), and N1-methyl-4-pyridone-3-carboxamide (4-PY). This implies that the higher the NAD+ level in the blood, the better the walking speed is maintained in older adults.
In addition to the improvement in physical function, the NMN group also showed significant improvements in sleep quality. Assessed through the Pittsburgh Sleep Quality Index (PSQI), the NMN group scored lower on "Daytime Dysfunction" and "Global PSQI" compared to the placebo group, suggesting that NMN intake may help improve sleep quality in the elderly. The study did not observe any adverse effects related to the test substance intake, further evidencing the safety of NMN.
The significance of this study lies in its support of previous findings that NMN has a positive impact on the walking speed of the elderly and provides confidence that long-term intake of NMN may have beneficial effects on the health of older adults. Walking speed is closely related to the risk of mortality and is an important indicator of physical function and daily living function in the elderly. Maintaining a faster walking speed helps older adults better preserve life functions, potentially extending healthy life expectancy. Therefore, exercise programs and medical interventions like NMN, which can help maintain walking speed, may contribute to extending the length of healthy life.
In summary, this study provides strong scientific support for the application of NMN in the field of elderly health, especially in increasing NAD+ levels, maintaining walking speed, and improving sleep quality. As the population ages, these findings are of great significance for developing new interventions to improve the quality of life for the elderly.
